An Experimental Study on the Transplantation of Oral Carcinoma


Abstract
The purpose of this study is to evaluate the possibilities of conservation of auto- and homo-transplanted, oral carcinoma experimentally induced by a chemical carcinogen through gross and histopathologic observation.
70 male ; Syrian hamsters were employed in this study, divided into 1 control and 3 experimental groups. .16 animals in control received no treatment. ,34 animals in experimental group 1 had the left pouch mucosa painted 3 times weekly with 0.5% 9,10-dimethyl1,2-benzanthracene (DMBA) in mineral oil.
Each 2 animals in control were sacrificed in 4, 6, 8, 10, 12, 14, 16, 20 weeks after experiments, and each 4 animals in experimental group I were sacrificed in 4, 6, 8, 10, 12, 14 weeks after experiments. The pouch mucosae of animals in control and experimental group were removed and routinely processed via hematoxylin-eosin. (H-E) staining for histopathologic observation.
Also the left tumorous pouch mucosae of 10 animals among experimental group 1 survived till 16 weeks during experiments were removed and divided into 4 pieces. Among above specimens 1 piece was routinely processed via H-E staining for histopathologic confirmation of carcinoma, and another 1 piece was autotransplanted into pouch mucosa of opposite side in same animal. Also another 1 piece was homotransplanted into pouch mucosa of another 10 Syrian hamsters of experimental group 2, and the other piece was homotransplanted into pouch mucosa of another 10 Syrian hamsters of experimetal group 3 which were injected intramuscularly into femoral muscle 3 times weekly with 5 mg/kg hydrocortisone sodium succinate (HSS) for 16 weeks.
After auto- and homo-transplantation the changes of transplanted site were grossly observed, and each ¢¥S animals in each experimental group were injected with HSS for 4 weeks under above conditions.
Also the specimens from all transplanted sites in all experimental groups were routinely processed via H-E staining for histopathologic observation. The obtained results were as follows:
1. In general hamster. pouch;, mucosae showed.¢¥.lustopathologic :features of epithelial dysplasia in 10 weeks and carcinoma in 12 weeks under application of 0.5% DMBA in mineral oil 3 times weekly.
2. In general the possibility of conservation of transplanted experimental hamster pouch carcinoma induced by DMBA was lower in homotransplantation than in autotransplantation into hamster pouch mucosa.
3. In general the possibility of conservation of homotransplanted experimental hamster pouch carcinoma induced by DMBA was higher in experimental group 3 ( pre-transplantation-injected with HSS for 16 weeks) than in experimental group 2(: not pre-transplantation-injected with HSS):
4. In general the possibility of conservation of transplanted experimental hamster pouch carcinoma induced by DMBA was not influenced by post-transplantation-injected with HSS for 4 weeks.
